Blood-Brain Barrier: A Comprehensive Review of Its Structure, Function and Role in CNS Diseases

Abstract The blood-brain barrier (BBB) is a highly specialized and dynamic structure that plays a crucial role in maintaining the homeostasis of the central nervous system (CNS). Composed of endothelial cells, pericytes, and astrocytic end-feet, the BBB regulates the exchange of molecules between the bloodstream and the CNS, thereby protecting the brain from harmful substances and maintaining its internal environment. Despite its importance, the BBB is often compromised in various neurological disorders, such as Alzheimer's disease, Parkinson's disease, and multiple sclerosis, leading to increased permeability and the infiltration of toxic substances into the brain. This review aims to provide a comprehensive overview of the structure, function, and regulation of the BBB, as well as its role in CNS diseases.
